Abstract Mature embryos from a large number of varieties were cultured on Linsmaier & Skoog medium. Plants regenerated from the callus obtained, together with their progeny, showed somaclonal variation in the greenhouse and field. Many plantlets were recovered from cells cultured on medium with 1.5% NaCl. Several selected plants showed even higher salt tolerance. A few cell lines survived at a sublethal NaCl concentration (1.75%). Adding Al2(SO4)3 to the medium reduced cell proliferation. Callus was also affected by other medium modifications required for Al solubilization. Some plants that were regenerated from callus selected on modified medium, with or without Al, expressed a degree of Al tolerance. Selection for cold tolerance was attempted by long-term culture of callus at sublethal temperatures (11-13 deg C). Some plants that were regenerated from cell selection and tested under hydroponic conditions showed improved cold tolerance. The progeny of plants regenerated for the different stresses were sent for field testing in Africa. Results so far confirm the value of somaclonal variation and in vitro selection for stress tolerance in breeding.
